Mr. Osomatsu originally started as a gag manga series called Osomatsu-kun by Fujio Akatsuka, which ran from 1962 to 1969. The original series followed the Matsuno brothers, who are all identical sextuplets who cause mischief.

The series is pretty faithful in its adaptation, following the six siblings: Karamatsu, Choromatsu, Ichimatsu, Jyushimatsu, Todomatsu and, finally, Osomatsu, who is the oldest brother and leader of the group. However, in the show, the brothers are ten years older and are in a more contemporary setting. Each episode follows the brothers through some hilarious everyday and sometimes outlandish scenarios.
